Mechanism of Action

The CoT fraction of human genomic DNA consists primarily of rapidly annealing, Interspersed Repetitive Sequences (IRS). Morphix CoT1 DNA is prepared from human placental DNA that has been sheared, denatured, and re-annealed under controlled conditions to isolate these repetitive fractions, including the Alu and Kpn families.
When incorporated into a hybridization reaction, Morphix CoT1 functions as a competitive blocker:

  1. Competitive Annealing: The enriched repetitive strands in the CoT1 DNA anneal to their complementary sequences (SINEs and LINEs) on the target genomic DNA.
  2. Specific Targeting: By effectively occupying these repetitive sites, the reagent prevents labeled probes or primers from binding non-specifically.
  3. Signal Optimization: This ensures that hybridization occurs only at unique target sequences, significantly lowering the background signal and increasing data clarity.
